This refers to a drive error... Make sure that you are not using a USB hard drive and that you're internal HD runs at 7200rpm. If you're internal drive is fast enough and you are not using a USB external, maybe try and repair disk permissions in disk utilities if you're using a Mac, not too sure how you do that on a PC. I suspect that if the 'shrieks' are harmonics, you'll only hear them when the wave is in a non-zero position, which is why it's audible only when it's positioned up or down. If it's directly in the center, there is no signal to distort; i.e., silence.

I have actually never had any problems with version 5.53 yet, other than Altiverb and Waves messing up my exits from the program, but I know from before that Nuendo at least has been very reluctant to handle samplerates not matching the project.
